You will always loose some. You probably lost that much because you used a big filter.
For such a small batch I'd rather use syringe filters or brew more gear.

The only thing I can think of for this loss is that I used a 250ml
Vacuum filter and some got lost in the filter. If that is it would a syringe filter work better for these types of small batches?
If loss is a normal, how much on average should I account for?
33% seems steep to me. Thanks so much
What kind of filtration system did u use? Wednesday size was it? What type of filter? (Pvdf, nylon, etc)

Surface area of filter has alot to do with it.
Would be less with syringe filter, yes.
Laboratory glass filtration systems where you install the sterile filter manually, have small surface area as well.

Many bottle top filters have larger filters, so slightly more loss.

Other factors: measuring with syringes or beaker will make you think you have a certain amount, but you actually have more or less.
Class A graduated cylinder would be the best method.

I’ve only had this issue once in my brewing, with masteron. I poured the unfiltered product into bottles, and filtered it later with another batch. Was about 20 ml that wouldn’t go thru

I think sometimes the filter just gets clogged. My vacuum pump simply wouldn’t pull one more ml thru that filter.

That is somewhat possible. Do you use a glass media bottle and a filtertop and hook it up to a vaccum pump?

I’m using Autofil, just like the OP. I use 500 ml instead of 250 ml. All plastic
