Forgot to mention, this was just after it was assembled, it was then moved into the "clean room" with a ceiling mount industrial HEPA filter
1) Tubing from VWR.....(NOT Home Depot)
2) Tubing was hermetically sealed into media jug caps.
3) Left Side Front 1000ml Media Jug Was Unfiltered Product. (after being on a hotplate for (x) amount of time
4) Left Side Rear 500ml Media Jug contained a heavier Carrier oil than what I used in my actual brew.
5) Right Side 1000ml Media Jug was filtered product.
6) With a Mec-O-Matic Dolphin 50 (I think that was the model) peristaltic set-up using a filter bell and when calculating tubing volume/filter bell volume, the unfiltered jug would be empty.
You never want to run a dry filter bell, it will "crush the filter membrane", so when the 1000ml jug was empty, I closed off the valve coming from unfiltered product, opened the heavier carrier oil valve. Turned the pump back on to " flush the remaining finished product through the filter bell until I ended up with the exact amount of finished product as I started with.
7) Immediately would attach a digital "DISPENSETTE III" to the finished 1000ml Media jug.
8) Removed 10ml vials from Tuttnauer autoclave along with stoppers (keeping everything sterile)
9) Set dispensette to 10.5ml and began the filling process, adding a stopper after one 10ml serum vial was filled.
10) Had "help" set the fliptop on using pneumatic crimper, label would be applied if required. If going to bulk then placed in tamper evident 7.5mm mylar bag, and sealed after every 100 serum vials were filled.
11) Outside of mylar bag, label with compound name, mfg date so there was never any question as to what was what.
12) Flushed entire system with 50/50 mix of specific carrier oil/BA (overkill I know)
That’s a nice setup. I would like to mention that it is unnecessary to pipe the filter into a media bottle if you have the hood/shroud thing on the bottom of the filters. I just set the filter on top of the media bottle.
As far as air pressure “crushing” the filter membrane, I can’t imagine air pressure being more destructive than hydraulic pressure. The membranes are backed up with support plates to prevent this anyway. Did you read this somewhere on GE’s website or literature?
Sourcing med grade tubing is all well and good but those John Guest fittings are the weak link and not likely rated for this use. Sterile IV rigs have plenty of diverter and stop valves. I’d go for one of those.
The CR pumps I use are digital and pretty accurate and easy to use with touch screens. Way better than medical grade pumps available out there. For our purposes anyway.

I totally agree with you "crushing" I guess that wasn't the best term I should have used. The bell filter membrane being a composite non woven construction, When there is no solution while it is in use, I guess you could say it almost "collapses" on itself like squeezing a sponge (but on a much smaller level). I didn't want to risk damaging the filter and having to replace it. I will look up the specs and what it says on "running while dry" and post it up when I find the exact manufacturers specs and operating instructions. Def not debating on what you pointed out, what you posted is excellent info as well.
At the end of the day, I felt confident that the finished product was as sterile as I could possibly get it to be with the given space/equipment available at that point in time.
I have no doubt you certainly know what you're talking about and probably love talking "shop" as much as I do. If...….and only if I were involved as much as I used to be things would be much different/upgraded. At the time, I was concerned more about open air exposure to the finished product and working in a "clean-room" type environment.
For me it was certainly a starting point and gives others some things to use as a reference if wanting to take things a bit further from just a hand brake pump and screw on vacuum type filters.

You’d love the clean room I built. Exchanges air out every 45 seconds. To that spec anyway, haven’t done air balance. Even with that, everything is done under laminar flow. Even cooking and mixing of orals. I’m fairly certain this is all lost on Mexicans as they just want cheap. Potency, truth in labeling and cleanliness falling low on priority. Even so, I still want to do all I can. I hate the thought of making someone sick.
you hit the nail on the head withat that statement, i couldnt even bare the thought of that!! I got really really really (did I mention really) sick from Mexican Brand "Animal Power" Cyp for my 1st cycle in my early 20s.

Youd prob remember the times were cattle implants (cant remember the name of em...snyovex maybe) that had test prop and had to use "Mr. T's Super Solvent" with the syringe filters? And finaplix cartridges??

I can only imagine what a set-up you have for a clean room just by the fact you use PLC controlled pumps, that stuck out to me big time right off the rip.
Things certainly have changed, for the better or worse....im not to sure yet. I havnt been "actively posting" on boards from the past 2 or so years and even in that short time the change has been astounding. (but then again...who am I to say anything) Everyone has their own reasons why they do what they do.
When you get to the stages of where you have a clean room even for your orals, Id say that whatever type of work you are doing you are CERTAINLY doing it right!!
